Abstract

The oxidation-bromination reaction of captopril by bromine has been used to develop three methods for the determination of captopril in bulk drug and tablet formulations. In the direct titrimetric method, the sample solution or tablet extract is acidified and titrated with bromate solution in the presence of excess of bromide using methyl orange as indicator, the end-point being indicated by the bleaching of red colour. The indirect titrimetric method involves the addition of a known excess of bromate-bromide reagent and back titration of the residual reagent iodometrically, after a specified time. The spectrophotometric method also involves the addition of known excess of bromate in the presence of excess of bromide and determination of unreacted bromate by measuring the increase in absorbance of the dye, methyl orange, at 510 nm. Titrimetry is applicable over 1-7 mg (direct) and 2-18 mg (indirect) range of captopril, whereas spectrophotometry permits the assay in the concentration range 0.25-2.0 mug ml(-1) with an apparent molar absoptivity of 7.08x10(4) 1 mol(-1) cm(-1) and the Sandell sensitivity of 3.07 ng cm(-2). The methods are simple, rapid and successful for the analysis of some pharmaceutical formulations.
