An extractive spectrophotometric method for the detn. of promethazine hydrochloride in either the pure form or in its pharmaceutical formulations is described. The method is based on the formation of violet color product with anthranilic acid in the presence of N-bromosuccinimide. The reaction probably proceeds via preliminary oxidn. of the phenothiazine nucleus to a phenothiazinyl radical followed by coupling of the anthranilic acid in C-3 or C-7 position. Beer's law is obeyed in the concn. range 2.5-30.8 μg/mL at the max. absorption of 555 nm. Optimum anal. conditions were detd. The produced chromogen is stable for >3 days at room temp. The results compare favorably with those of official methods. Also, the method is specific for C-2 unsubstituted promethazine hydrochloride. [on SciFinder(R)]
