Abstract

An extractive spectrophotometric method for the detn. of promethazine hydrochloride in either the pure form or in its pharmaceutical formulation is described. The method is based on the formation of red colored product with 4-aminobenzoic acid in the presence of N-bromosuccinimide. The reaction is thought to proceed via preliminary oxidn. of the phenothiazine nucleus to a phenothizinyl radical followed by coupling of the 4-aminobenzoic acid in C-3 or C-7 position. Beer's law is obeyed in the range of concn. of 5-36.5 μg/mL at the max. absorption of 510 nm. Optimum anal. conditions were detd. The produced chromogen is stable for more than three days at room temp. Anal. data for detn. of the pure compd. is presented together with the application of the proposed method to anal. of some pharmaceutical prepns. The results compare favorably with those of official methods. Furthermore, the method is specific for C-2 unsubstituted promethazine hydrochloride. The possible reaction mechanism is suggested.
