The alkaline protease enzyme present in the mid gut of silkworm, Bombyx mori L. is the most important enzyme responsible for the conversion of mulberry protein to silk protein and its activity varies from breed to breed, between hybrids; and season to season, Among the two productive breeds, CSR2 expressed higher Protease activity and total midgut protein in all the three stages and seasons when compared to CSR4 and their hybrid CSR2 × CSR4 expressed higher enzyme activity and higher protein content when compared to their parents in all the three stages of collection and seasons. Similar results were recorded in robust breeds also, CSR18 excelled over CSR19 for both the enzyme and total midgut protein and their hybrid excelled over both the parents for enzyme activity as well as the midgut protein content. Among the hybrids, robust hybrid expressed better heterosis and heterobeltiosis for the total midgut protein content in all the three seasons reflecting the superiority of CSR18 × CSR19 for its productivity traits. It can be concluded from the present findings that the highest midgut protein content on 3rd day of 5th instar expresses the maximum conversion of mulberry protein to silk protein and the cocoons produced after spinning will have higher silk content.
