Two simple, accurate and precise methods for the determination of five phenothiazine drugs are described. The titrimetric method involves direct titration of drugs with N-bromosuccinimide (NBS). The spectrophotometric method is based on the oxidation of the drugs with a known excess of NBS and estimating the unreacted NBS with metol and sulphanilic acid. The stoichiometry of the reaction that forms the basis for titrimetry was assessed. Different variables affecting the colour formation in spectrophotometry were studied and optimised. At the maximum absorption of 530 nm, Beer's Law is obeyed in the range 0-40 mug ml(-1) The molar absorptivity and Sandel sensitivity of the produced charge-transfer complex with each drug were calculated. A proposal for the reaction mechanism is also presented. The proposed methods were applied to the analysis of pharmaceutical preparations containing these drugs. Statistical treatment of the experimental results indicates that the procedures are precise and accurate. Excipients used as additives in pharmaceutical formulations did not interfere in the proposed procedures. The reliability of the methods was established by parallel determination against the official BP methods.
