A simple and sensitive spectrophotometric method for the estimation of certain aromatic vic-diols like dopamine hydrochloride, levodopa, methyldopa, adrenaline, pyrocatechol, eugenol and guaiacol in either the pure form or in its pharmaceutical formulations is described. The method is based on the formation of a yellow to orange coloured product with semicarbazide hydrochloride in the presence of potassium persulfate. The reaction is thought to proceed via oxidation of aromatic vic-diols to an o-benzoquinone derivative followed by coupling with semicarbazide. Beer's law of the coloured species is obeyed in the range of 0.1-24 mug ml(-1) at maximum absorption (460-470 nm). The molar absorptivity was found to be in the range of 8.1 x 10(3) -2.62 x 10(4) l mol(-1) cm(-1) and the coloured species were stable for 30-60 min. All measurements were carried out in an alkaline medium at a room temperature. The method is highly reproducible and specific for these selected aromatic vie-diols. The common excipients used as additives in pharmaceuticals do not interfere in the proposed method, Analytical data for the determination of the pure compound is presented together with the application of the proposed method to the analysis of some pharmaceutical formulations. The results compare favourably with those of official methods and reported method.
